      目的 设计一种烟用接装纸中淀粉含量的测定方法。方法 选用糖化酶对烟用接装纸中的淀粉进行酶解,采用3,5-二硝基水杨酸(DNS)比色法对酶解产物葡萄糖含量进行测定,进而计算出烟用接装纸中的淀粉含量。研究在单因素试验分析的基础上,采用响应面分析法优化酶解法测定烟用接装纸中淀粉含量的最佳条件,建立数学模型,并对方法进行精密度和加标回收率的测试。结果 得到了酶解法测定烟用接装纸中淀粉含量的最佳条件,糖化酶用量为2.64 mg/mL,反应时间为35 min,反应温度为53 ℃,方法相关系数为0.999,淀粉检出限为1.1 mg/L,相对标准偏差为1.02%,加标回收率为95.2%~105%。结论 该方法操作简便,准确度高,适合于烟用接装纸中淀粉含量的测定。

      The work aims to design a method for determining the starch content in tipping paper for cigarette. Glucoamylase was used to degrade the starch in the tipping paper for cigarette. The glucose of the enzyme hydrolysis product in the solution was measured by 3,5-dinitrosalicylic acid colorimetry (DNS colorimetry) and the starch content in tipping paper for cigarette was calculated. Based on the results of the single factor experiments, the response surface methodology was used to optimize the optimal conditions of starch content determination in tipping paper for cigarette with enzyme hydrolysis and establish the mathematical model. Meanwhile, the precision and recovery rate of the method were tested. The optimal determination conditions of starch in tipping paper for cigarette were glucoamylase dosage of 2.64 mg・mL–1, reaction time of 35 min, reaction temperature of 53 ℃, correlation coefficient of 0.999, detection limit of starch of 1.1 mg・L–1, relative standard deviation of 1.02% and recovery rate of 95.2%-105% respectively. The method is easy to operate, with high accuracy and it can be used to determine the starch content in tipping paper for cigarette.
